Competitive pressure is forcing organizations to continually increase screening throughput. LEADseeker™ Multimodality Imaging System was developed to meet these demands by combining excellent detection with the ability to image whole plates in less than one minute. For screens such as luminescence cell-based assays, this means imaging plates faster without sacrificing detection sensitivity, a feature which has been described previously. This can drastically reduce the time required to quantitate all wells in a plate compared to traditional luminometers, particularly at higher densities (such as in 384 and 1536-well plates).
This poster describes the assessment and characterization of defined optical filters used for the luminescence mode of LEADseeker together with the DiscoveRx™ "HitHunter™ range of EFC chemiluminescent assays and the BRET2™ luminescence technology. For the DiscoveRx range, the Caspase 3 and p38 MAPK binding assay kits (GE Healthcare) were used as model systems with LEADseeker equipped with the supplied luminescence filter. For the BRET2 technology, the demo kit (PerkinElmer) was utilized and two new filters were defined for use with LEADseeker.
