The intracellular translocation of the EGFP-SMAD2 fusion protein can be monitored using fluorescence microscopes in the presence of an appropriate agonist such as transforming growth factor-b1 (TGF-b1), thereby providing a means of measuring SMAD2 activity and interrogating the TGF-b1 signalling pathway. The degree of activation of the TGF-b1 signaling pathway and SMAD2 can be quantified by measuring the cytoplasmic to nuclear ratio of fluorescence of the EGFP-SMAD2 fusion protein in different cellular contexts.
