Hints when charging Chelating media with Zn2+
.
As a general rule, more metal can be bound to the Chelating Sepharose media as the pH increases from 5.5 to 8.5. This, however, has to be considered in combination with the fact that the higher pH can cause metal hydroxide precipitates, and for Zn2+, it is not advisable to use basic conditions during loading.
When Zn2+ is used, formation of a precipitate on the addition of Na2CO3 to the eluent indicates saturation of the column, and can give an idea of the amount of Zn2+ that has bound.
