PreDictor 96-well filter plates for high-throughput process development

PreDictor™ 96-well filter plates are prefilled with BioProcess™ chromatography media for ion exchange and antibody affinity chromatography.

PreDictor plates support high-throughput process development (HTPD) by allowing parallel screening of chromatography conditions for binding, wash, and elution.
They can also be used for determination of adsorption isotherms.

PreDictor plates can be operated in either a manual or an automated workflow.

Assist software supports the PreDictor workflow from set-up of experimental design to data evaluation.

Data generated using PreDictor plates show good correlation with data from chromatography columns, making the plates an excellent tool for initial screening of process conditions.

PreDictor plates and Assist software shorten time-to-clinic and improve productivity by:

  • Decreasing experimental time from weeks to hours
  • Reducing sample consumption
  • Increasing process understanding through exploring an enlarged experimental space

Assist software has been developed to support PreDictor™ plates and the high-throughput methodology.

It guides the user through the workflow by suggesting an experimental design based on the type of study and experimental conditions. It also manages and evaluates the data generated.

PreDictor 96-well filter plates for high-throughput process development

Automated workflows provide a high degree of robustness and allow hand-off operation. PreDictor™ plates are a standard size, 1: 127.8 × 85.5 × 30.6 mm and fit into any equipment in automated workflows that accommodates this size of plate.

When using a robotic system for automated operation, the robot must be equipped to support the individual steps in the PreDictor protocol.

PreDictor 96-well filter plates prefilled with BioProcess™ chromatography media are fully tested for parallel screening of chromatographic conditions on Tecan Freedom EVO® workstation. The combination of Tecan’s robotic integration and liquid handling expertise together with the versatile PreDictor platform gives the user a fully automated system for high-throughput process development (HTPD).

Example scripts based on PreDictor protocols have been developed and are ready to be implemented on Freedom EVO.

To ensure robust and reliable processing of PreDictor plates on Freedom EVO, dedicated accessories for PreDictor are available from Tecan.

Screening plates
PreDictor AIEX(Anion exchange) screening plate 2μl/6μl contains the following chromatography medium volumes per well: Capto Q 2 μl, Capto DEAE 2 μl, Q Sepharose Fast Flow 6 μl and Capto adhere 6 μl. PreDictor AIEX screening plate 20 μl contains 20 μl per well of the corresponding media.

PreDictor CIEX(Cation exchange) screening plates 2μl/6μl contains the following chromatography medium volumes per well: Capto S 2 μl, SP Sepharose Fast Flow 6 μl and Capto MMC 6 μl. PreDictor CIEX screening plate 20 μl contains 20 μl per well of the corresponding media.

PreDictor HIC screening high hydrophobicity contains following chromatography media:
Phenyl Sepharose 6 Fast Flow (low sub), Capto Butyl, Phenyl Sepharose 6 Fast Flow (high sub) and Capto Phenyl (high sub).

PreDictor HIC screening low hydrophobicity contains following chromatography media:
Butyl-S Sepharose 6 Fast Flow, Octyl Sepharose 4 Fast Flow, Butyl Sepharose 4 Fast Flow and Capto Octyl.
