Technical Information
MegaBACE™ Long Read Matrix, with linear polyacrylamide (LPA), produces long read lengths with tremendous accuracy. It enables average read lengths in excess of >500 bases, with PHRED accuracy scores of greater than 20 (in fact, when used in conjunction with a slower, three-hour run time, read lengths of up to 1000 bases are possible). In genotyping applications, MegaBACE™ Long Read Matrix is optimized for resolutions of 0.3 bp using MegaBACE™ ET400-R Size Standards; 0.5 bp using MegaBACE™ ET550-R Size Standards, and 2 bp using MegaBACE™ ET900-R Size Standards.
MegaBACE™ Long Read Matrix, with linear polyacrylamide, contributes to long read lengths and high accuracy. A plasmid sample was sequenced using DYEnamic™ ET Dye Terminator Kit (MegaBACE™). Excess salts and dye terminators were removed using ethanol precipitation. The sample was electrokinetically injected into the capillary and separated via electrophoresis at 4 kV for 300 min. The results show a Cimarron Base-caller read length of 1001 bases and a PHRED 20 score of 959.
