Technical Information
Multiple injection of SNuPe™ reactions on MegaBACE™ DNA Analysis System. Three SNuPe™ reactions were injected into two different capillaries. illusta AutoSeq™ 96 was used for post-reaction cleanup.
MegaBACE™ SNuPe™ Genotyping Kit enables four-color SNP genotyping of PCR products with MegaBACE™ DNA Analysis System. The kit contains Thermo Sequenase™ DNA polymerase and four dye-labeled dideoxynucleotides (terminators) in a single-tube premix format.
SNP genotyping on the MegaBACE™ takes advantage of the instrument's flexibility to load twelve, 96-well sample plates over a period of approximately 30 min, using a proprietary method of repeated, time-spaced injections. A multiple injection marker is added to each sample prior to injection, acting as an internal control and as a demarcation of one injection from the next. A total of 1200 SNP genotypes can be obtained with a turnaround time of less than 2 h.
MegaBACE™ SNP Profiler v1.0 is a Windows™ based software that automatically processes signal data and outputs SNP genotypes. Allele calling is based on the allele signal intensity in each channel and allele mobility in the specific injection. The software enables project management ability. Multiple runs of the same SNP or multiple injection run data can be analyzed at the same time.
