Deep Purple™ Total Protein Stain
Technical Information
Deep Purple™ Total Protein Stain* is an ultra-sensitive, rapid, and easy-to-use fluorescent stain for the detection of proteins in gels or blots following electrophoretic separation. Less than 0.5 ng protein per band can be detected in 1-D dilution series in gels and less than 1 ng protein per band when blotted onto nitrocellulose or PVDF membranes. Deep Purple™ Total Protein Stain is used to stain both 1-D and 2-D gels in both backed and unbacked format, and has been shown to have high compatibility with mass spectrometry. Deep Purple™ Total Protein Stain is also used to stain proteins blotted onto nitrocellulose or PVDF membranes, and has been validated for use with the ECL Plex™ Western blotting system. Because the stain is free from heavy metals, it can be safely disposed after use.
Deep Purple™ Total Protein Stain can be used with ultraviolet transillumination (365 nm), light boxes (400-500 nm), laser-based scanners (457, 488, or 532-nm excitation), and CCD cameras.
Deep Purple™ Total Protein Stain is available in standard and bulk pack sizes. The standard pack is sufficient for staining two large-format gels (e.g., large-format, DALT Gel 12.5 2-D gels) or 20 mini-gels. The bulk pack is sufficient for staining 10 large-format gels or 100 mini-gels.

Two-dimensional protein gel stained with Deep Purple™ Total Protein Stain. Protein sample consists of HBL100 breast cell line and BT474 breast cell carcinoma. First dimension: Immobiline™ DryStrip pH 3-10 NL, 24 cm run on Ettan™ IPGphor IEF System. Second dimension: DALT Gel 12.5 run on Ettan™ DALTtwelve Large Vertical Electrophoresis System. Scanning: Typhoon™ 9410 Variable Mode Imager: Excitation, green laser (532 nm); emission 560LP filter, PMT 530 V, 100 µm resolution. The gel image shows the pH 3-8 region, where most proteins are present.
Low molecular weight markers in two-fold dilutions from approximately 128-1 ng, blotted onto Hybond-P (PVDF). The blot was stained with Deep Purple™ Total Protein Stain according to the standard protocol. The detection limit was 1 ng or better.
