SOURCE™ Media/RESOURCE™ and Tricorn Columns for HIC

  • A range of high-resolution HIC media designed for low- to medium-pressure systems ranging from a peristaltic pump to ÄKTA™design systems and FPLC™ System.
  • Ideal for final polishing in a purification process.
  • High performance at low back pressure, even with high sample loads of up to 25 mg of protein.
  • Features ether (SOURCE™ 15ETH), isopropyl (SOURCE™ 15ISO) and phenyl (SOURCE™ 15PHE) hydrophobic ligands coupled to the 15 μm monodispersed rigid polystyrene/divinylbenzene matrix.
  • Prepacked 1-ml RESOURCE™ columns for fast and high-resolution separation are available individually in each of the three matrices or as a set in the RESOURCE™ HIC Test Kit.
  • SOURCE 15PHE 4.6/100 PE is a Tricorn™ column designed for high resolution at lab scale and for optimization studies when scaling up.

The figure on the right shows four runs in an optimization series of eight runs to find the optimal running conditions for the purification of protein tyrosine phosphatase on SOURCE 15PHE 4.6/100 PE (Tricorn™). In the optimization series, the concentration of ammonium sulfate, gradient length, pH, flow rate and additives were varied. The activity was measured by p-nitrophenyl phosphate (pNPP) activity assay at 405 nm. In Run 8, the increase in concentration of ammonium sulfate from 1.5 M to 2.0 M and the increase of the length of the gradient resulted in a better separation.
