MultiTrap™ 96-well filter plates for high-throughput expression and purification screening

MultiTrap™ 96-well filter plates are prefilled with chromatography media for tasks such as screening GST-tagged and His-tagged proteins, purifying antibodies, enriching proteins by immunoprecipitation (immunoaffinity), sample preparation and desalting.

MultiTrap supports rapid, high-throughput screening by allowing parallel screening of conditions to determine the optimal solubility and expression levels of target proteins. The plates can also be used to optimize purification conditions for binding, wash and elution. Parallel small-scale sample preparation also has applications in biomarker research and screening in the discovery phase of drug development.

A wide range of chromatography products based on MultiTrap is available. Plates can be operated either manually or in automated liquid-handling workstations. Optimization data generated using MultiTrap plates show good correlation with data from chromatography columns. The conditions identified can then be used in larger scale studies using prepacked columns and ÄKTAdesign™ systems such as ÄKTAxpress™.

Automation
Automated workflows provide a high degree of robustness and allow hands-off operation. MultiTrap plates are standard-sized (127.8 × 85.5 × 30.6 mm) and can be used in any automated equipment that accommodates this format.

MultiTrap filter plates prefilled with affinity chromatography media have been fully tested in the parallel screening of target proteins and chromatographic conditions on Tecan Freedom EVO® workstation and Hamilton MicroLab STAR™. Results showed excellent reproducibility.

The combination of Tecan’s robotic integration and liquid-handling expertise, together with the versatile MultiTrap platform, gives users a reliable and fully-automated system for early, high-throughput screening of target proteins for biomarkers, drug lead generation and lead identification.

Example scripts based on MultiTrap protocols have been developed for both vacuum and centrifugation and are ready to be implemented on Freedom EVO.
