Reversed Phase Chromatography

Reversed Phase Chromatography (RPC) separates molecules according to differences in their hydrophobicity. RPC has become increasingly important for high-resolution separation and analysis of proteins, peptides and nucleic acids. The technique is ideal for applications such as peptide mapping or purity checking and is often used for final polishing of oligonucleotides and peptides. Separations can be performed using isocratic elution; jpwever, most frequently, gradient elution is used in order to minimize the run time. RPC can also be used to separate molecules of significantly differetd hydrophobicity using step elution.
