Ion Exchange chromatography

Ion exchange chromatography is among the most precise and frequently used methods for the fractionation of biological substances. Biomolecules with even small differences in charge can be separated. Very high resolution is obtained by choosing the optimal ion exchanger and the optimal separation conditions.

GE Healthcare offers a complete range of ion exchange gels, all hallmarks of our trusted quality and performance. Choose from a variety of columns prepacked for your convenience, or purchase bulk media and pack your own column.

MiniBeads™ columns - Prepacked Tricorn™ and PC columns

MiniBeads are based on a nonporous, monodispersed matrix of rigid, hydrophilic particles, substituted with quaternary amino (Q) or methyl sulfonate (S) groups. The very small particle size (3 µm), uniformity and physical rigidity of the particles create ideal conditions for extremely high resolution ion exhange separateions at relatively high flow rates and low back pressures. The strong ion exchange groups (Q and S) maintain their charge over a broad pH range, allowing selection of the most suitable pH for each application. MiniBeads ion exchange media are suitable for purification and analysis of microgram-milligram quantities of proteins, peptides or oligonuclotides at a very high resolution.

Mini Q™ and Mini S™ are available in Tricorn high-performance columns and in Precision Columns (PC) for high-resolution micro-purification.

MonoBeads™ columns - Prepacked Tricorn and PC columns

MonoBeads ion exchange chromatography media are based on a hydrophilic matrix made from mondispersed (10 µm), rigid, polystyrene/divinyl benzene porous particles, substituted with strong ion exchange groups Q (quaternary amino) and S (methyl sulfonate).

The small particle sieze allow fast binding and dissociation to facilitate high resolution while the uniformity of the particles ensures high flow rates at low back pressures. Mono™ Q and Mono™ S are used for purification of milligram quantities of proteins, peptide and oligonucleotides at high resolution.

Mono Q and Mono S are available in Tricorn™ high performance columns and in Precision Columns (PC) for high-resolution micro-purification.

SOURCE ™ 15 and SOURCE 30 - Prepacked Tricorn, RESOURCE columns and Lab packs

SOURCE ion exchange chromatography media are based on a hydrophilic matrix made from monodispersed, rigid, polystyrene/dininylbenzene and substituted with the two strong ion exhange groups Q (quaternary amino) and S (methyl sulfonate). SOURCE media are available in two particle sizes, 15 µm and 30 µm.

The 15 µm particle, SOURCE 15, for rapid, high-resolution purification and is ideal for final polishing in a purification process.

SOURCE 30 (30 µm) is an alternative to SOURCE 15 for large-scale applications where speed rather than resolution is a priority.

SOURCE 15 Q and SOURCE 15 Q are available in labpacks as well as prepacked in Tricorn™ high-performance columns and in RESOURCE™ columns for rapid separations.

SOURCE 30 Q and SOURCE 30 S are available in lab packs.

Sepharose™ High Performance - Prepacked HiTrap , HiLoad and Lab packs

Q Sepharose High Performance (HP) and SP Sepharose HP are 34 µm media suitable for intermediate or polishing steps that require high resolution. Both chromatography media are available in lab packs as well as prepacked in convenient HiTrap columns. When increased capacity is required, use prepacked HiLoad columns.

Capto™ - Prepacked HiTrap , HiScreen columns and Lab packs

Capto ion exchange media are based on a highly rigid agarose base matrix that offers
outstanding pressure/flow properties with optimized pore structure. Capto media are intended for general use in large-scale bioprocess operations.

Capto S, Capto Q and Capto DEAE are, respectively, strong cation, strong anion and weak anion exchange media for packed bed chromatography. They increase speed and throughput in capture and intermediate purification.

Capto ViralQ is specifically intended for the purification of viruses.

Capto MMC is a multimodal cation exchanger. The multimodal functionality gives a different selectivity compared with traditional ion exchangers. This includue binding of proteins in the presence of high salt concentrations.

Capto adhere is a stron anion exchanger with multimodal functionality giving a different selectivity compared to traditional anion exchangers. Capto adhere has been designed for post Protein A purification of monoclonal antibodies.

All Capto media are available in lab-packs. The different media are also available in HiTrap columns as well as in the HiTrap™ Capto Selection kit, to rapidly screen for the most suitable medium for a specific application. Prepacked HiScreen™ columns with the different Capto media are designed for process development work.

Sepharose™ Fast Flow - Prepacked HiTrap , HiPrep columns and Lab packs

Sepharose Fast Flow is based on a matrix of particles with a mean size of 90 µm. The medium is well suited for capture or intermediate purification and in large-scale purifications.
Sepharose Fast Flow matrices are substituted with a range of ion exchanger groups giving the opportunity to test and use different selectivities. The strong ion exchangers Q and SP maintain their charge over a broad pH range, allowing selection of the most suitable pH for each application.

Ion exchangers containing weak ion exchange groups (DEAE, CM and ANX) offer alternative selectivitites, but over a narrower pH working range.

All chromatography media are available in lab-packs. The different media are also available in HiTrap™ columns as well as in the HiTrap IEX Selection kit, to rapidly screen for the most suitable medium for a specific application. When increased capacity is required, use prepacked HiPrep™ columns

Sepharose™ XL - Prepacked HiPrep , HiTrap columns and Lab packs

Sepharose XL chromatography media are based on a matrix of 90 mm particles, made from 6% agarose and highly cross-linked for chemical and physical stability, substituted with quarternary ammonium (Q) or sulfopropyl (SP) groups. The ionic groups are bound to long, flexible dextran chains that have been coupled to the agarose. This increases the exposure of the Q or SP groups thereby raising the binding capacity to a very high level.

Q Sepharose XL virus licensed is specifically intended for the purification of viruses.

All chromatography media are available in lab-packs. Q and SP Sepharose XL are also available in HiTrap™ columns. When increase capacity is required, use prepacked HiPrep™ columns
