Amersham™ ECL™ Prime Western blotting detection reagent Summary The latest addition to the Amersham ECL family, Amersham ECL Prime, is at least twice as sensitive as Amersham ECL Plus, with a limit of detection (LOD) in the low picogram range. The reagent is characterized by greatly increased signal stability, which allows repeated exposures and makes it easier to process several blots in one experiment. In addition, the 3- to 5-fold increase in intensity of the signals emitted by Amersham ECL Prime means that proteins may be detected using primary and secondary antibodies at 3-fold higher dilutions than with Amersham ECL Plus, contributing to lower background and reducing the consumption of costly antibody reagents. Western Blotting - Principles and Methods Summary Western blotting, also known as immunoblotting, is a well established and widely used technique for the detection and analysis of proteins. The method is based on building an antibody:protein complex via specific binding of antibodies to proteins immobilized on a membrane and detecting the bound antibody with one of several detection methods. The Western blotting method was first described in 1979 and has since become one of the most commonly used methods in life science research. The aim of this handbook is to guide and inspire beginners as well as experts toward successful Western blotting. It provides expertise and support, taking you through the complete Western blotting workflow, from sample preparation to detection and analysis. Chapters 2 to 8 follow the Western blotting workflow step by step and describe theoretical as well as practical aspects of the technique together with useful hints and tips. Examples of typical applications as well as some new approaches to Western blotting are described in Chapter 9. The enhanced oxidation of luminol is a multi-step reaction regulated by the HRP amount. Use of IMAC magnetic beads for simplified detergent screening for optimization of purification workflows of histidine-tagged membrane proteins Summary Integral membrane proteins are main targets for development of new pharmaceuticals. Affinity purification is a powerful tool that has become the prime choice as capture step in membrane protein purification. Recombinant membrane proteins are commonly purified by use of polyhistidine tags and immobilized metal affinity chromatography (IMAC). The choice of detergent is the major key factor for successful solubilisation and further purifications of integral membrane proteins to avoid protein loss and inactivation. Therefore a detergent screen is often necessary to find the optimal detergent for each purpose. The use of the Membrane Protein Purification Kit allows for rapid, small-scale screening of optimal purification conditions for histidine-tagged membrane proteins. The kit contains IMAC magnetic beads pre-charged with Ni2+-ions (His Mag Sepharose™ Ni), selected detergents, buffers and an easy-to-follow protocol. Close
