We offer a range of kits for template preparation and manual sequencing. Sequencing is based primarily on Sequenase Version 2.0 T7 DNA polymerase, a widely used enzyme for radioactive DNA sequencing. T7 Sequencing Kit can be used to read through difficult regions of secondary structure and is a good choice for sequencing double stranded DNA. PCR Product Pre-Sequencing Kit enables rapid, enzymatic PCR product clean-up prior to sequencing.
Manual DNA sequencing is a cost-effective method for sequence analysis of relatively few samples and/or when relatively short sequences are required. Sanger sequencing reactions include radiolabeled dideoxynucleotide triphosphates to give fragments that are separated on denaturing polyacrylamide gels and analyzed using autoradiographic film or a phosphorimager plate. Emphasis is placed on convenience in template preparation and efficient polymerases that can generate sequences from a range of templates, even those with GC-rich regions. This demands reagents that generate even band intensities, tolerate substrate analogs well, and can be used to read through difficult regions of secondary structure. Reagent mixes can be prepared specifically to give either long or short reads.
