DNA and RNA synthesis play a central role in molecular biology. Synthetic oligonucleotides (or oligos) are used as primers for sequencing, PCR and cloning, probes for arrays, etc. They also have applications in antisense therapy whereby the oligonucleotide modifies the expression of key genes in a disease pathway. This demands large-scale synthesis of extremely high-quality oligonucleotides. Oligosynthesis may even include modified bases to add new functionalities such as resistance to enzymatic degradation.
The synthesis of oligonucleotides must be efficient and accurate to avoid an exponential increase in errors in the molecular population after 20, and especially over 100 incorporations of nucleosides. Once synthesized, the product must be cleaned up chromatographically to remove incomplete molecules and other side products of the process. The oligo is then checked for purity, normally by a combination of liquid chromatography and mass spectrometry.
