The task of cleaning, setup, and sterilization of a fixed bioreactor typically takes one working day and requires copious amounts of high purity water and energy. The installation of a single-use bioreactor takes minutes. Disposable bioreactors have a small footprint and eliminate the need for laborious cleaning and cleaning validation.
WAVE Bioreactor Systems have revolutionized upstream operations by making cell culture faster, flexible, and more cost-effective. Cells and culture medium only come in contact with a presterilized, disposable culture chamber called a Cellbag Bioreactor that is placed on a rocking platform to induce waves.
The gentle wave motion prevents settling and provides oxygenation without bubbles. WAVE Bioreactor Systems have been used extensively for monoclonal antibody production as well as for cultivation of anchorage-dependent cells on various microcarriers. WAVE Bioreactor Systems are available in a variety of scalable configurations for suspension, microcarrier, batch, fed-batch, or perfusion culture.
Cellbag Bioreactors are made from clear, multilayer laminated film for bioinert fluid contact and high mechanical strength. The disposable bioreactors are available in sizes for volumes from 250 ml to 500 l for use with the WAVE Bioreactor System.
Cellbag Bioreactor has needleless ports that allow sterile access into and out of the bag and single-use accessories such as tubing, connectors, and clamps are available for fluid handling. Gas exchange is possible via 0.2 μm air filters. There are also optical sensors available for optimal process control. These include accurate single-use pH sensors or reusable oxygen probes.
Mixing operations can be monitored and controlled to suit the application using WAVE Mixer. The instrument makes it possible to mix materials contained in disposable bags in a completely sterile manner. The rocking platform induces a wave motion in the liquid without an impeller or other invasive mixer. The WAVE Mixer has been optimized for efficient mixing and applications include thawing, pooling, mixing, and media and buffer preparation.
