ImageQuant LAS 4000 is a digital imaging system for sensitive, quantitative imaging of gels, blots, and colonies. ImageQuant LAS 4010 Fully equipped imager that in addition performs ultraviolet (UV) and visible (RGB) fluorescence imaging.
- Exceptional sensitivity for quantitation of biomolecules labeled with chemiluminescent probes, and expandable to multifluorescent UV, visible, and IR imaging. Allows gel documentation of Coomassie and silver-stained samples
- Accurate quantitation of closely spaced bands in complex protein samples for gels and films up to 21 x 14 cm (25 x 25 cm using the optional lens). Low noise for longer exposure times and lower background, important for precise quantitation of very weak signals
- Wide linear dynamic range for precise quantitation of faint and dark bands over four orders of magnitude in the same image, avoiding time consuming multiple film exposures
- Distortion, dark frame, and flat frame corrections are applied to each imaging mode for optimal precision and uniform quantitation
- Sensitive detection of less than 40 pg of protein on a Western blot. Sensitivity may be increased by "binning" of up to 8 x 8 pixels and smoothing
- Chemiluminescent and colorimetric signals can be captured in the same image without changing the lens. Optional multifluorescence detection extends functionality to the capture of fluorophores such as Cy2, Cy3, and Cy5 and Deep Purple Total protein stain
ImageQuant LAS 4000 is designed to quantitate the highly sensitive detection of proteins using Amersham ECL Western blotting reagents. A free combination of light sources and filters can be incorporated into the imager according to your needs.
The system consists of an intelligent dark box, camera, lens, and automated four-position filter wheel with a standard filter for EtBr and open positions for the filters delivered with the optional fluorescent light sources. Focusing, filters, illuminators, and exposure time are remotely controlled by a computer. Simple operation allows adjustment without having to open the cabinet. The intelligent light box provides automation and user safety. UV light sources are housed inside the cabinet, and can only be switched on when the door is closed.
A bright, wide aperture FUJINON™ F0.85 lens, specially developed for chemiluminescence imaging, projects sharp images onto a patterned CCD chip. By diagonally aligning octogonal pixels, the gap between pixels in the horizontal and vertical planes is reduced. This pattern allows an effective image resolution of 6.3 Mpixels.
The user-friendly control software has several capture modes for achieving optimal sensitivity and dynamic range including increment, repetition, and program modes. For image analysis, advanced and flexible ImageQuant TL software is available separately and comprises a dedicated suite of analysis applications to provide high levels of automation and accuracy in analysis of gels, blots, and colony counting.

Amersham ECL Select
Summary
Amersham ECL Select is a highly sensitive reagent for chemiluminescent Western
blotting detection. It is the most sensitive chemiluminescent detection reagent
in the Amersham ECL product family. The dynamic range of Amersham ECL Select is
complementary to that of Amersham ECL and Amersham ECL Prime. The outstanding
signal intensity of Amersham ECL Select makes it suitable for the most
demanding Western blotting applications including the detection of minute
protein quantities. You can perform multiple exposures because of the long
lasting signals of Amersham ECL Select and the convenient time window between
experiment and analysis.

Amersham™ ECL™ Prime Western blotting detection reagent
Summary
The latest addition to the Amersham ECL family, Amersham ECL Prime, is at least
twice as sensitive as Amersham ECL Plus, with a limit of detection (LOD) in the
low picogram range. The reagent is characterized by greatly increased signal
stability, which allows repeated exposures and makes it easier to process
several blots in one experiment. In addition, the 3- to 5-fold increase in
intensity of the signals emitted by Amersham ECL Prime means that proteins may
be detected using primary and secondary antibodies at 3-fold higher dilutions
than with Amersham ECL Plus, contributing to lower background and reducing the
consumption of costly antibody reagents.

Western Blotting - Principles and Methods
Summary
Western blotting, also known as immunoblotting, is a well established and
widely used technique for the detection and analysis of proteins. The method is
based on building an antibody:protein complex via specific binding of
antibodies to proteins immobilized on a membrane and detecting the bound
antibody with one of several detection methods. The Western blotting method was
first described in 1979 and has since become one of the most commonly used
methods in life science research.
The aim of this handbook is to guide and inspire beginners as well as experts
toward successful Western blotting. It provides expertise and support, taking
you through the complete Western blotting workflow, from sample preparation to
detection and analysis. Chapters 2 to 8 follow the Western blotting workflow
step by step and describe theoretical as well as practical aspects of the
technique together with useful hints and tips. Examples of typical applications
as well as some new approaches to Western blotting are described in Chapter 9.
Moreover, protocols and recipes are provided in Chapter 11, including a
recommended standard Western blotting procedure, to help you design and run
your experiments. Over the past decade, improved detection methods and software
have brought quantitative analysis to Western blotting and this handbook
provides examples and protocols to help you adapt your methods and obtain more
quantitative data from your Western blots. Finally, a troubleshooting guide is
provided in Chapter 10, just in case!

which cover topics closely connected to Western blotting, such as sample
preparation or 2-D gel electrophoresis. Where appropriate, we refer to these
